Ishtar is the guardian deity of uruk which is gilgamesh's capital
Gilgamesh doesn't need the key because he can access his vaults anyway
a normal human can't use the key because they lack the authority anyways
now since ishtar is the deity of the capital she has enough authority to use the key to seal the vaults (can't take from it though). Any other god that doesn't have ties to gil or uruk wouldn't be able to do it.
Also IIRC ishtar cursed gil so that if he reunites with enkidu she would be there to mess them up because they messed with her during their lives (gods be petty and all)
